چکیده

Rhizoctonia muneratii ATCC 13247 was capable of catalyzing the conversion of 25-hydroxycholesterol to the 3-oxo-4-ene. Hydroxylation at the 6alpha and 6beta positions with the concomitant 3-oxo-4-ene transformation was also observed. The addition of Triton X-100 to the growth medium immediately before steroid addition selectively prevented the appearance of the respective 6alpha-hydroxylated products. The observed conversions were indicative of the utility of microbial transformations in preparing hydroxylated steroids which can be useful for further synthetic modification or for study as pharmacological agents.

Metabolism of 27-, 25- and 24-hydroxycholesterol in rat glial cells and neurons.

The metabolism of 27-, 25- and 24-hydroxycholesterol in cultures of rat astrocytes, Schwann cells and neurons was studied. 27- and 25-Hydroxycholesterol, but not 24-hydroxycholesterol, underwent 7 alpha-hydroxylation with subsequent oxidation to 7 alpha-hydroxy-3-oxo-delta 4 steroids in all three cell types. When cells were incubated for 24 h with 0.28 nmol of 27-hydroxycholesterol in 10 ml of ...

Interferon regulator factor 1/retinoic inducible gene I (IRF1/RIG-I) axis mediates 25-hydroxycholesterol-induced interleukin-8 production in atherosclerosis.

AIMS In this study, the role of retinoic inducible gene I (RIG-I)-mediated signalling in the inflammation of atherosclerosis was investigated to explain the pathology of atherosclerosis. METHODS AND RESULTS Human and mouse primary cells were exposed to 25-hydroxycholesterol followed by examination of gene expression and activation of the signal pathway with biochemical and molecular biologica...

Transformation of Potato (Solanum tuberosumcv.Savalan) by Chitinase and β-1,3-Glucanase Genes of Myco-Parasitic Fungi Towards Improving Resistance to Rhizoctonia solani AG-3

Potato (Solanum tuberosum L.) an agro-economically important food crop in the world, is sensitive to many fungal pathogens including Rhizoctonia solani (AG-3), the causal agent of stem and root rot diseases. Chitinase and glucanase are cell wall degrading enzymes which have been shown to have high antifungal activity against a wide range of phytopathogenic fungi. In the present study, plasmid p...

Biotransformations of putative phytoecdysteroid biosynthetic precursors in tissue cultures of Polypodium vulgare.

Incubation of calli and prothalli of Polypodium vulgare with different tritium-labelled ecdysteroids has led to modification of some previous assumptions about the biosynthesis of ecdysteroids in plants. Thus, 25-deoxy-20-hydroxyecdysone was transformed efficiently in both tissues into 20-hydroxyecdysone (20E), but no 25-deoxyecdysteroids such as pterosterone and inokosterone were formed. Likew...

Kinetics of G1 progression in 3T6 and SV-3T3 cells following treatment by 25-hydroxycholesterol.

The kinetics of cell cycle progression in continuously proliferating 3T3 fibroblasts and two tumor transformed derivatives (3T6 and SV 3T3 cells) following treatment by growth-factor deprivation (serum starvation) or 25-hydroxycholesterol were studied. Normal 3T3 cells were found to respond immediately (in the first cycle) to growth factor deprivation by leaving the cell cycle from G1, whereas ...
